RGT Project 1

This project will give you experience in optimization of a common protocol, bacterial transformation with a defined plasmid.

There are a number of methods that can be used to insert a plasmid into a bacterial cell. Select one method and then define the optimal transformation conditions that gives the highest transformation efficiency for that method.

Transformation efficiency is defined as the number of transformed cells per microgram of plasmid DNA.

You will have 4 weeks to perform you experiments and write the journal style paper.

You will exchange papers for an inclass peer-review session on February 27th.

You will submit your revised paper to your instructor on March 1st.

You will be given a plasmid and XL1Blue E. coli cells upon submission of an experiment and request for materials.
